The analysis of vitamin D status, with special emphasis on 25-hydroxyvitamin D and 1,25-dihydroxyvitamin D, is gaining interest in clinical studies due to the classical and non-classical effects attributed to this prohormone. In this research, the influence of the two steps preceding determination (viz. sample collection and preparation) on the quantitative analysis of vitamin D and its more important metabolites has been studied. Two preparation approaches, deproteination and solid-phase extraction (SPE), have been evaluated in terms of sensitivity to delimit their application, thus establishing that detection of 1,25-dihydroxyvitamin D cannot be addressed by protein precipitation. Concerning sample collection, serum and plasma reported high accuracy (above 83.3%) for vitamin D and metabolites, while precision, expressed as relative standard deviation, was below 12.9% for all analytes in both samples. Statistical analysis revealed that serum and plasma provided similar physiological levels for vitamin D3, 24,25-dihydroxyvitamin D3 and 25-hydroxyvitamin D3, while significantly different levels were obtained for 1,25-dihydroxyvitamin D3, always higher in plasma than in serum. Sample collection and treatment have proved to be significant in the analysis of vitamin D and its relevant metabolites.  相似文献

Microwrinkles were formed by treating poly(dimethylsiloxane) (PDMS) surfaces with a mixture of concentrated sulfuric acid and nitric acid at 80 °C followed by a water rinsing step. The acid treatment oxidized the treated area, which also swelled owing to absorption of water during the rinsing step. This absorption of water induced a compressive stress on the PDMS surface that caused the formation of wrinkles. The appearance of wrinkles was controlled by the time interval between the end of the acid treatment and the beginning of the water rinsing step. Wrinkles were formed when the time interval was shorter than 5 s, as water was absorbed by the hygroscopic acid molecules within the treated surface. For longer time intervals of 300 s, this phenomenon was not observed. Based on these findings, a procedure for forming wrinkles in specific areas was developed by hydrating the chosen areas with wet filter paper cut into specific shapes after the acid treatment. The formation of wrinkles was confined to the area under the piece of paper. The orientation and alignment of the wrinkles at the boundaries were also characterized. © 2014 Wiley Periodicals, Inc. J. The compatibility of multiple functions at a single interface is difficult to achieve, but is even more challenging when the functions directly counteract one another. This study provides insight into the creation of a simultaneously multifunctional surface formed by balancing two orthogonal functions; water repellency and enzyme catalysis. A partially fluorinated thiol is used to impart bulk hydrophobicity on the surface, and an N‐hydroxysuccinimide ester‐terminated thiol provides a specific anchoring sites for the covalent enzyme attachment. Different ratios of the two thiols are mixed together to form amphiphilic self‐assembled monolayers, which are characterized with polarization‐modulation infrared reflection–absorption spectroscopy and contact angle goniometry. The enzyme activity is measured by a fluorescence assay. With the results collected here, specific surface compositions are identified at which the orthogonal functions of water repellency and enzyme catalysis are balanced and exist simultaneously. An understanding of how to effectively balance orthogonal functions at surfaces can be extended to a number of higher‐scale applications.  相似文献

In this paper, a parameter identification method to determine surface shortwave fluxes using temperature and thickness measurements of sea ice in CHINARE 2006 is presented. Adopting a new standard for the calculation of the thermodynamic properties of seawater named TEOS‐10, the surface shortwave fluxes are calculated by the temperature and thickness observations that were measured at Nella Fjord around Zhongshan Station, Antarctica. New simulations for temperature profiles in a different measurement period are performed by three parameterization schemes including the present method, Zillman and Shine. All numerical results are compared with in situ measurements. Results show that better simulations of the surface shortwave radiations and temperature distributions are possible with the identification method than Zillman and Shine. Therefore this method is valid, and the obtained shortwave radiation function can be applied in sea ice modeling. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

A facile click chemistry method of immobilizing surface‐functionalized polymer vesicles on casted polymeric PAN substrates is described. Microporous PAN membranes were subjected to hydrochloric acid hydrolysis to obtain surface carboxylates. The carboxylic groups were activated with EDC/NHS‐solution and were then reacted with propargylamine to introduce alkyne groups for CuAAC reactions. The alkyne functionality of the modified membrane surface was verified by reaction with an azide functional click dye both before and after the immobilization of azide‐functionalized ABA vesicles. The efficient postfunctionalization of the membrane with alkyne allowed quantitative coverage of the membrane surface with a polymersome monolayer, as confirmed by immobilization of polymerzomes loaded with a fluorescent dye. Polymersome monolayers immobilized on alkyne functionalized PAN‐membranes were characterized by cryo‐SEM and monolayers were confirmed by atom force microscopy. We report an improved method for the preparation of trichloromethylcarbinols from enolizable ketones. Trichloromethylcarbinols were obtained in good to excellent yields by using of a combination of CHCl3, n-BuLi, and chlorotitanium (IV) triisopropoxide. In this follow-up of our previous work [30], the author proposes a high-order semi-implicit method for numerically solving the incompressible Navier–Stokes equations on locally-refined periodic domains. Fourth-order finite-volume stencils are employed for spatially discretizing various operators in the context of structured adaptive mesh refinement (AMR). Time integration adopts a fourth-order, semi-implicit, additive Runge–Kutta method to treat the non-stiff convection term explicitly and the stiff diffusion term implicitly. The divergence-free condition is fulfilled by an approximate projection operator. Altogether, these components yield a simple algorithm for simulating incompressible viscous flows on periodic domains with fourth-order accuracies both in time and in space. Results of numerical tests show that the proposed method is superior to previous second-order methods in terms of accuracy and efficiency. A major contribution of this work is the analysis of a fourth-order approximate projection operator.  相似文献
